Critical parameters of the sample application are:

  • The precise position of the sample: Chromatography must start simultaneously for all samples on a plate to ensure comparability of RF of separated zones. For that, a standardized plate layout is adopted.
  • The application volume: Precise application volumes are prerequisite for qualitative and quantitative analysis. A common problem is sample overloading, which decreases the separation power and causes tailing of zones.
  • No damage to the layer: Damages to the layer during sample application can affect the movement of the mobile phase. Do mark the application position on the layer.
